Whole cell extracts (15 µg total protein) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.5 µg/mL of Purified anti-B-Raf (clone W21141A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

Description

B-Raf is one of three proteins in the RAF family of serine/threonine protein kinases. B-Raf regulates the mitogen-activated protein kinase/extracellular signal-regulated kinase (MAPK/ERK) pathway. Like the other two RAF kinases, A-Raf and C-Raf, B-Raf relays extracellular signals to the nucleus through the MAPK/ERK pathway and regulates cell division, differentiation, and secretion. RAF proteins share similar protein domains and motifs, including multiple protein binding domains such as Ras, lipid, and the 14-3-3 binding motif. RAF proteins mainly reside in the cytoplasm in an inactive state by binding to 14-3-3 proteins. RAFs translocate to the membrane compartment by activation and binding with RAS proteins. Similar to other RAF kinases, B-Raf phosphorylates MAP2K1 (MEK) proteins to activate the MAPK/ERK pathway. Mutations in the gene which encodes B-Raf are highly correlated with inherited cardiofaciocautaneous syndrome and various types of cancers. The B-Raf V600E mutation results in a constitutively active kinase leading to aberrant cell proliferation and invasion. Such mutations are linked to hairy cell leukemia, melanoma, colon cancer, and non-small-cell lung cancer. B-Raf inhibitors are considered as anticancer targets for metastatic melanoma, kidney cancer, and liver cancer.
