IL-4 is a pleiotropic cytokine that is produced by activated T cells mast cells and basophils. IL-4 elicits many different biological responses, but has two dominant functions. The first is regulating differentiation of naïve CD4+ T cell to the Th2 type. Th2 cells produce IL-4, IL-5, IL-10 and IL-13, which tend to favor a humoral immune response while suppressing a cell-mediated immune response controlled by Th1 cells. The second is regulating IgE and IgG1 production by B cells. The 8D4-8 antibody can neutralize the bioactivity of natural or recombinant IL-4.

Each lot of this antibody is quality control tested by ELISA assay. For sandwich ELISA capture applications, a concentration range of 1.0 - 8.0 µg/ml is recommended when used in conjunction with biotinylated MP4-25D2 (Cat. No. 500803) as the detection antibody, and recombinant human IL-4 (ELISA Std.) (Cat. No. 571809) as the protein standard. It is recommended that the reagents be titrated for optimal performance for each application.

Due to the possible complexities and variations of uses of biofunctional antibodies in different assays and because of the large product portfolio, BioLegend does not currently perform functional assays as a routine QC for the antibodies. However, we do provide references in which the antibodies were used for functional assays and we do perform QC to verify the specificity and quality of the antibody based on our strict specification criteria.
