Total lysates (15 µg protein) from Daudi (Human, Positive control), Jurkat (Human, Negative control) and A20 (Mouse) were resolved by electrophoresis (4-20% Tris-glycine gel), transferred to nitrocellulose, and probed with 1:1000 (0.5 µg/ml) purified anti-POU2AF1 antibody, clone 2E9C01. Proteins were visualized using chemiluminescence detection by incubation with HRP Goat anti-Mouse secondary antibody (Cat. No. 405306, 1:3000 dilution). Direct-Blot™ HRP anti-β-actin antibody was used as a loading control (Cat. No. 643807, 1:8000 dilution).

Description

POU2AF1, also known as OBF1, is a B lymphocyte specific transcriptional coactivator. POU2AF1 plays important regulatory roles in the function and maturation of B cells. It stimulates transcription of the immunoglobulin genes by interaction with Oct-1 or Oct-2. The POU2AF1 gene encodes a single mRNA that produces two isoforms, p34 and p35, by post-translational processing. Both isoforms are expressed at similar levels and are able to stimulate octamer-dependent transcription. The cytoplasmic p35 isoform of POU2AF1 regulates pre-B cell proliferation and BCR signaling through interacting with the tyrosine kinase Syk. POU2AF1 deficient mice have shown reduced numbers of circulating B cells in the bone marrow and failed to form germinal centers in the spleen.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 10.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

The POU2AF1 gene encodes a single mRNA that produces two isoforms, p34 and p35. This antibody recognizes both.
