Total cell lysates (15 µg protein) from Molt-4, HeLa and NIH3T3 were resolved by 4-20% Tris-glycine gel electrophoresis, transferred to nitrocellulose, and probed with 2 µg/mL purified anti-MYDGF (SF20) antibody (clone W16077A) antibody (upper). Proteins were visualized using a goat anti-rat-IgG secondary antibody conjugated to HRP and chemiluminescence detection. Direct-Blot™ HRP anti-β-actin Antibody was used as a loading control (lower). Lane M: MW ladder.

Total lysates (15 µg protein) from 293T and 293T/MYDGF CRISPR/Cas9 knockout (KO) cells were resolved by electrophoresis (4-12% Bis-Tris gel), transferred to nitrocellulose, and probed with 1:250 diluted (2 µg/mL) purified anti-MYDGF (SF20) antibody, clone W16077A (upper) or 1:3000 diluted anti-GAPDH antibody (lower). Proteins were visualized by chemiluminescence detection using a 1:3000 diluted goat anti-rat-IgG secondary antibody conjugated to HRP for anti-MYDGF (SF20) antibody, and a donkey anti-rabbit IgG Antibody conjugated to HRP for GAPDH. Lane M: Molecular weight ladder. Non-specific bands were observed over 150 kD in the 293T WT and KO cell lines tested.

HeLa cells were fixed with 4% paraformaldehyde (PFA) for 15 minutes, permeabilized with 0.5% Triton X-100 for 3 minutes, and blocked with 5% FBS for 60 minutes. Then the cells were intracellularly stained with 0 and 2 µg/ml anti-MYDGF (SF20) antibody (clone W16077A) overnight at 4° followed by Alexa Fluor® 594 (red) conjugated goat anti-rat IgG for one hour at room temperature. Nuclei were counterstained with DAPI (blue). The image was captured with a 60X objective.

Description

Myeloid-derived growth factor (MYDGF), also known as Stromal cell-derived growth factor SF20, is encoded by an open reading frame on chromosome 19 (C19orf10) and is secreted by bone marrow-derived monocytes and macrophages. MYDGF protects cardiomyocytes from apoptosis in a PI3K/AKT-dependent signaling pathway and promotes angiogenesis in endothelial cells through a MAPK1/3, STAT3, and CCND1-mediated signaling. During myocardial infarction, MYDGF therapy has been found to increase angiogenesis in the infarcted area and attenuate infarct size, scar formation, cardiac dilatation, and systolic dysfunction. In comparison to wild-type mice, MYDGF-deficient mice developed larger infarcts and had more severe systolic dysfunction.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.2 - 2.0 µg per ml. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Clone W16077A does not cross-react with mouse (in-house tested). As per the Human Protein Atlas RNA level dataset, Molt-4 is expected to have a low level of MYDGF expression.
