Proteins were visualized using an HRP goat anti-mouse IgG (minimal x-reactivity) antibody and chemiluminescence detection.

    A549 cells were fixed with 4% paraformaldehyde (PFA) for ten minutes and then were permeabilized with 0.5% Triton X-100 for 5 minutes followed by blocking with 5% FBS for 30 minutes. Then the cells were intracellularly stained with 5 µg/mL anti-FOXA2 (clone P87H4B7) in blocking buffer overnight at 4°C and followed by DyLight™ 594 (red) conjugated goat anti-mouse IgG for one hour at room temperature. Actin filaments were labeled with Alexa Fluor® 488 Phalloidin (green). Nuclei were counterstained with DAPI (blue). The image was captured with a 60X objective.

Description

FOXA2 belongs to the Forkhead box family. This transcription factor is also known as Hepatocyte Nuclear Factor 3 beta (HNF-3β). It is involved in mouse embryonic development, differentiation and maturation of endoderm derived organs, and glucose and lipid metabolism. Loss of FOXA2 has been connected to diseases such as hyperinsulinemic hypoglycemia and lung adenocarcinoma metastasis.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 1.0 - 5.0 µg/ml. For immunocytochemistry, a concentration range of 4.0 - 10 μg/ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
