Total cell lysates (15 µg total protein) from HepG2, A431 HEL and Raw264.7 cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with Purified anti-MyPt1 Antibody, clone Poly19251, at a 1:10000 dilution overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP donkey anti-rabbit IgG Antibody (Cat. No. 406401) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker.

Description

Myosin phosphatase target subunit 1 (MYPT1) is a regulatory subunit of protein phosphatase 1. It regulates the interaction of actin and myosin downstream of the guanosine triphosphatase Rho. The GTP-bound, active form of RhoA (GTP.RhoA), through its interaction with myosin-binding subunit (MBS) of myosin phosphatase, regulates the phosphorylation of myosin light chain (MLC). Rho inhibits myosin phosphatase through the action of Rho-kinase. Four splice-variant isoforms for the MYPT1 gene have been identified.

Store at -20°C. Upon initial thawing, apportion into working aliquots and store at -20°C. Avoid repeated freeze-thaw cycles to prevent denaturing the antibody. For long-term storage, keep the antibody at -80°C.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 1:10000 - 1:20000 dilution. It is recommended that the reagent be titrated for optimal performance for each application.

The non-catalytic subunit of myosin phosphate, the myosin binding subunit (MBS), is called M130 or MYPT1.
