Experimental Design

Application:	Cell Culture
Cells used:	Lineage negative Bone Marrow (CD3e-, Ly-6C/G-, CD11b-, CD45R/B220, TER-119-)
Brief Protocol:	1. Isolate bone marrow 2. Remove RBC with lysis buffer 3. Deplete Lineage+ cells with magnetic depletion 4. Count and suspend in mem-α containing 20% FBS, mSCF 50ng/mL, mIL3 10ng/mL , mIL6 20ng/mL (R&D), mIL7 25ng/mL (R&D) 5. Add 5000 cells/well (24 well plate) 6. Infect with lentivirus MOI=100 and polybrene=6 ug/mL
Results Summary:	Cell viability was equal to current recombinant murine IL-3 from Peprotech.
