Brief Protocol: |
The s+16a clone specifically blocks the ecto-enzyme ARTC2 on mouse T cells when injected 15min prior to sacrificing the mouse (50µg i.v.). This prevents ADP-ribosylation of T cell surface proteins including the P2X7 ion channel. ADP-ribosylation of P2X7 occurs at 4°C, however, when isolated T cells are brought back to 37 °C this post-translational modification induces opening of the ion channel subsequently inducing cell death. |
Results Summary: |
Injection of Treg-Protector preserves the vitality of freshly prepared Tregs retaining their suppressive capacity e.g. during in vitro suppression assays. |
