Cell suspensions from mouse spleen (works really well) or blood (less efficient). |
Brief Protocol: |
We isolate a spleen from C57BL/6 mice, and prepare a cell suspension. After centrifugation cells are resuspended in 500 ul of RBC lysis buffer (1X). We centrifuge again immediately, and resuspend cells for counting. For FACS staining we usually put up to 5 x 10^6 cells in a 50ul volume and add a mix of fluorescent anti-mouse antibodies in FACS buffer for 1h at room temperature. |
Results Summary: |
While lysis of red blood cells from spleen requires only one wash with 500ul of RBC, lysis of blood cells requires at least 3 incubation in 500 ul (15 min each with small agitation). This product is very mild. |
