Cell suspensions from mouse lymphoid tissues such as spleen and lymph nodes. |
Brief Protocol: |
We isolated cells from peripheral lymphoid tissues, pooled spleen and lymph nodes from C57BL/6 mice, and FACS-sorted CD27- gd T cells. The CD27- gd T cells were stimulated overnight with IL-1b and IL-23, and then 4h with PMA and ionomycin for 4h at 37C. Brefeldin A was added for the last 2h of re-stimulation. Then we performed an intracellular FACS staining using anti-mouse IL-22-PE antibody. We usually put up to 5 x 10^6 cells in a 50ul volume and add anti-mouse IL-22-PE diluted at 1/100 final in permeabilization buffer for 1h at room temperature. |
Results Summary: |
Anti-mouse IL-22-PE antibody provides a strong signal that allows reliable staining to study gd T cells. |

Figure shows results from CD27- gamma delta T cells purified from spleen and lymph node of wild type C57BL6 mice and stimulated overnight with IL-1b+IL-23 and then re-stimulated for 4h with PMA/Iono. Cells were stained with anti-IL-22-PE and anti-IL17A-FITC. |
