Brief Protocol: |
We isolated cells from tumour implanted in from C57BL/6 mice, by using collagenase DNase treatment, and stained them with Zombie Violet dye to exclude dead cells. We put up to 5 x 10^6 cells in a 50ul volume and add Zombie Violet diluted at a 1/1000 final in PBS for 15min at 4C. |
Results Summary: |
Zombie Violet provides a bright signal that allows strong and reliable staining to exclude dead / dying cells, from various types of organs from mouse. |
Additional Notes: |
Our laboratory has interest in tumour-infiltrating immune cells and we have tested many different products to exclude dead cells after collagenase DNase treatment. We choose to use routinely Zombie Violet dye as this was the best for our combination with more 9 other colours.
Figure shows zombie violet against CD45 staining from E0771 tumour cell suspension of C57BL6 mouse.
