Group 2 Innate Lymphoid Cells and GATA3
BioLegend is pleased to continually provide reagents for new and exciting fields of study. Innate lymphoid cells (ILCs) are an emerging population of cells with key roles in innate immunity, tissue repair/remodeling, epithelial cell integrity, and microorganism defense. Recently, a proposal was made to classify these cells based on their transcription factor and surface marker expression, rather than their variable cytokine production. The most well known ILCs are natural killer (NK) cells and lymphoid tissue-inducer (LTi) cells, which have recently been classified as Group 1 and Group 3 ILCs, respectively. In a study by Hoyler et al., Group 2 was established by their expression of GATA3, which is commonly thought of as a Th2 transcription factor.

Group 2 ILCs (ILC2) were mainly identified by cell surface markers (i.e., IL-7R?, Sca1, CD117, ICOS)  and Type 2 cytokines like IL-5 and IL-13. However, transcription factors vital for ILC2 maintenance were not established. Hoyer et al. found that ILC2 express GATA3, a transcription factor typically associated with Th2 cells. They also identified a precursor population in the bone marrow, termed LSIG (Lin-Sca-1hiId2hiGATA3hi) or ILC2 Precursors (ILC2P). GATA3 was found to be constitutively expressed in both ILC2 and IL2CPcells and absolutely required for their function and development. ILC2P could be converted into ILC2 with IL-25 and IL-33 treatment. Adoptive transfer of ILC2P cells into Rag2 Il2rg mice promoted eosinophil recruitment in the lungs and mucin production from goblet cells and angiogenin expression by paneth cells in the intestines, leading to helminth parasite expulsion and control.
