Cytokine release syndrome (CRS) is a systemic inflammatory condition characterized by the activation of immune cells leading to excessive production of inflammatory cytokines. Triggers of CRS include CAR-T cell therapy, sepsis, and other factors. The resulting cytokine feedback loop can induce severe tissue damage. A critical step in CRS research involves an efficient and consistent quantification of the local immune mediators. Thus, obtaining high-quality tissue lysates is crucial for such analysis.
Sample lysis can be a time-intensive process depending on the chosen method. Traditional sonication, while effective, processes one sample at a time and requires thorough cleaning between samples. In contrast, bead mill homogenization allows the simultaneous processing of multiple samples with high reproducibility, producing lysates suitable for downstream applications.
In this webinar, we present a workflow for tissue lysate preparation using the Omni Bead Ruptor Elite™ bead mill homogenizer and the simultaneous quantification of 13 key CRS biomarkers using the LEGENDplex™ Cytokine Release Syndrome panel. These advances provide valuable insights into the cellular mechanisms of CRS, fostering the development of targeted immunotherapies.
