Description

IL-27, a heterodimer protein, belongs to the IL-12 cytokine family. IL-27 is composed of the Epstein-Barr virus-induced gene 3 (EBI-3), a 34 kD glycoprotein that is related to the p40 subunit of IL-12; and p28, a 28 kD glycoprotein that is related to the p35 subunit of IL-12 and p19 subunit of IL-23.

IL-27 triggers expansion of antigen-specific naïve CD4+ T cells, promotes polarization of Th1 cells, and inhibits the development of Th2 and Th17 cells. It is known that IL-27 is an early product of activated antigen-presenting cells (APCs). IL-27 plays an important role in regulating the activity of B and T lymphocytes.

Human CD33 signal peptide (Met1-Ala16), mouse EBI3 (NM_015766, Tyr19-Pro228), and mouse p28 (NM_145636, Phe29-Ser234) joined via a GGGSGGGSGGGTGGGS linker, expressed in 293E cells.
Molecular Mass
The total predicted molecular mass of mouse IL-27 is ~49 kD. The non-reduced and the DTT-reduced proteins migrate as one band at approximately 55 kD by SDS-PAGE.
Purity
>95%, as determined by Coomassie stained SDS-PAGE, prior to lyophilization.
Formulation
Lyophilized in sterile-filtered PBS, pH 7.2, containing 1% BSA, 0.09% sodium azide, and protease inhibitors.

Storage & Handling
Upon receipt, store unopened vials between 2°C and 8°C immediately and use within 12 months from date of receipt. Prior to use, reconstitute the lyophilized powder with 0.2 mL of PBS containing a carrier protein (e.g., 1% BSA, protease free), pH 7.4. Re-cap vial, vortex. Allow the reconstituted standard to sit at room temperature for 15 minutes, vortex again to mix completely. The reconstituted standard stock solution can be aliquoted into polypropylene vials and stored at -70°C for up to one month. Do not re-use diluted standards. Use a manual defrost freezer and avoid repeated freeze thaw cycles.

ELISA - Quality tested

Recommended Usage

Each lot of this protein is quality control tested by ELISA assay. For use as an ELISA standard, a standard curve comprised of two-fold dilutions from 5 ng/mL to 0.078 ng/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

To measure mouse IL-27, this recombinant protein can be used as a standard in a sandwich ELISA format, when used in conjunction with the purified MM27-7B1 antibody (Cat. No. 516902) as the capture antibody and the biotinylated B02P6E6 antibody (Cat. No. 525904) as the detection antibody.

Note: For measuring mouse IL-27 in serum, plasma or cell culture supernatant, LEGEND MAX™ Mouse IL-27 Heterodimer ELISA Kit with Pre-coated Plates (Cat. No. 438707 & 438708) are specially developed and recommended.

A heterodimeric cytokine consisting of EBV-induced gene-3 (EBI3, an IL-12 p40-related protein) and p28 (a newly discovered IL-12 p35-related protein).
Distribution

Produced by antigen-presenting cells, including macrophages and dendritic cells.

Function
Drives rapid clonal expansion of naïve CD4+ T cells, promotes Th1 polarization and IFN-γ production; plays a role in the anti-inflammatory and antitumor reponses, and has potential in autoimmune disease therapy.
