Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of purified anti-RAP1 antibody (clone W18011A) overnight at 4°C.     Whole cell extracts (250 µg total protein) prepared from Raji cells were immunoprecipitated overnight with 2.0 µg of purified rat IgG2b, κ isotype control antibody (Cat. No. 400602) (panel A) or purified anti-RAP1 antibody (clone W18011A) (panel B). The resulting IP fractions and whole cell extract input (6%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane and probed with a rabbit control antibody against a separate epitope of RAP1.     Raji cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with Triton X-100 for 10 minutes, and blocked with 5% FBS for 60 minutes. Cells were then intracellularly stained with 5 µg/mL (1:100 dilution) of either purified rat IgG2b, κ isotype ctrl antibody (Cat. No. 400602) (panel A) or purified anti-RAP1 antibody (clone W18011A) (panel B) overnight at 4°C, followed by incubation with Alexa Fluor® 594 goat anti-rat IgG antibody (Cat. No. 405422) at 2.0 µg/mL (1:250 dilution). Nuclei were counterstained with DAPI and the image was captured with a 60X objective.

Description

Repressor/activator protein 1 (RAP1), also called telomeric repeat-binding factor 2-interacting protein 1 (TERF2IP), is one of six subunits of the shelterin complex, which regulates telomerase activity and protects telomeric DNA through the generation of t-loops. RAP1 forms a heterodimer with shelterin subunit TRF2 and regulates telomere length, recombination, and transcription. This complex also functions in the repression of homology-directed repair (HDR) and prevents telomere deletions. In the cytoplasm, RAP1 functions as an inhibitor kinase (IKK) adaptor and modulator of the nuclear factor- κB (NF-κB) pathway as well as an activator of NF-κB transcriptional activity. TERF2IP S205 phosphorylation has been implicated in the regulation of activation, apoptosis, and senescence of endothelial cells.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.125 - 1.0 µg/mL. For immunoprecipitation, the suggested use of this reagent is 2.0 µg/test. For immunocytochemistry, a concentration of 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone does not recognize mouse RAP1.

ICC testing was conducted on Raji cells fixed with 4% paraformaldehyde and permeabilized with either Triton X-100 or methanol.  Both methods were compatible with RAP1 staining.
