Whole cell lysates (15 µg) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane and probed with a 1.0 µg/mL (1:500 dilution) of purified anti-RNA Polymerase II RPB5 antibody (clone 3Y11) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Ponceau S solution was used to show protein loading (lower). Lane M: Molecular weight marker.

Description

DNA-dependent RNA polymerase catalyzes the transcription of DNA into RNA using the four ribonucleoside triphosphates as substrates. The common components of RNA polymerases I, II, and III synthesize ribosomal RNA precursors, mRNA precursors, and many functional non-coding RNAs and small RNAs, such as 5S rRNA and tRNAs, respectively. Pol II is the central component of the basal RNA polymerase II transcription machinery. Pols are composed of mobile elements that move relative to each other. In Pol II, RPB5 is part of the lower jaw that surrounds the central large cleft and is believed to grab the incoming DNA template. It seems to be the major component in this process.
