C57BL/6 mouse frozen liver section was fixed with 4% paraformaldehyde (PFA) for 10 minutes at room temperature and blocked with 5% FBS for 30 minutes at room temperature. Then the section was stained with 10 µg/ml of anti-mouse CLEC4F (clone 3E3F9) Alexa Fluor® 647 (red) and 10 µg/ml of anti-mouse CD105 (clone MJ7/18) Alexa Fluor® 594 (green) overnight at 4°C. The image was captured by 20X objective.

CLEC4F, also known as Kupffer cell receptor or KCR, is a 80 kDa, type II transmembrane glycoprotein member of the C-type lectin superfamily. CLEC4F is expressed on resident Kupffer cells and infiltrated monocytes in the liver, bu not expressed on macrophages in other tissues. The biological function of CLEC4F has not been elucidated. Recent report showed that CLEC4F interacts with alpha-galactosylceramide (α-GalCer) in a calcium-dependent manner and participates in the presentation of α-GalCer to natural killer T (NKT) cells.

Each lot of this antibody is quality control tested by immunohistochemical staining on frozen tissue sections. For immunohistochemistry, a concentration range of 2.5 - 10 µg/ml is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
