Experimental Design

Application:	Primary Cell Culture, Lentivirus infection
Cells used:	Lineage depleted (CD3e-, Ly-6C/G-, CD11b-, CD45R/B220, TER-119-) Bone marrow
Brief Protocol:	1. Isolate bone marrow 2. Remove RBC with lysis buffer 3. Deplete Lineage+ cells with magnetic depletion 4. Count and suspend in mem-α containing 20% FBS, mSCF 50ng/mL, mIL3 10ng/mL (Biolegend), mIL6 20ng/mL (R&D), mIL7 25ng/mL (R&D) 5. Add 5000 cells/well (24 well plate) 6. Infect with lentivirus MOI=100 and polybrene=6 ug/mL.
Results Summary:	For BM-Lin- cells, mSCF from BioLegend can support the cell survival and proliferation comparably with mSCF from Peprotech.
Additional Notes:	Reduced cost and virtually no difference in cell number (n=3). We have switched to BioLegend mSCF for future use in similar experiments.
