Brief Protocol: | We isolated cells from spleen and peripheral lymph nodes of C57BL/6 mice and purified CD4 T cells by negative selection. Cells were polarized on CD3 precoated 24-well plates in the presence of anti-mouse CD28 (clone 37.51), anti-mouse IL-4, anti-mouse IFN-γ, mouse IL-6, human TGF-β1 and mouse IL-23 for 3 days. We restimulated cells with PMA/ionomycin and analysed IL-17 expression by intracellular cytokine staining using PE anti-mouse-IL-17A. |
Results Summary: | Polarization conditions induce robust expression of IL-17A in a large fraction of CD4 T cells. |
