Anti-mouse CD1d Biotin for Flow Cytometry

Good option to stain CD1d.
Experimental Design

Application: Flow cytometry
Cells used: Mouse splenocytes
Brief Protocol: Resuspend in PBS/BSA and count the cells. Load up to 5x106 cells onto a V-Bottom 96-well-plate. Spin down, resuspend in 50uL of PBS/BSA containing αFcγR (2.4G2) 30 µg/ml and incubate 15min at 4°C. Spin down and resuspend in 50 µl antibody staining solution containing anti-CD1d Biotin (1:400). Incubate 15min at 4°C in the dark. Wash with PBS/BSA, spin down and resuspend in 50 µl staining solution containing SA-fluorescent labeled. Wash twice with PBS/BSA, resuspend and transfer to FACS tubes to read. Use PI to exclude the dead cells.
Results Summary: CD1d is expressed by marginal zone B cells in the spleen.
