Experimental Design |
Application: | Flow cytometry |
Cells used: | Mouse splenocytes |
Brief Protocol: | Resuspend in PBS/BSA and count the cells. Load up to 5x106 cells onto a V-Bottom 96-well-plate. Spin down, resuspend in 50uL of PBS/BSA containing αFcγR (2.4G2) 30 µg/ml and incubate 15min at 4°C. Spin down and resuspend in 50 µl antibody staining solution containing anti-CD8 Brilliant Violet 510™ (1:200). Incubate 15min at 4°C in the dark. Wash twice with PBS/BSA, resuspend and transfer to FACS tubes to read. Use PI to exclude the dead cells. |
Results Summary: | CD8 is expressed by CD8 T cells in the spleen. |
