Subsets of macrophage and B cells in the peritoneum express CD73.

Experimental Design

Application:	Flow cytometry
Cells used:	Peritoneal exudate
Brief Protocol:	Peritoneum of two C57Bl/6 mice were washed with 10ml room temperature PBS 0.6mM EDTA, and placed directly on ice. Cells were then pooled, resuspended, and stained in PBS 1% BSA 2mM EDTA. Cells were blocked for a minimum of 10 min on ice with αFcγRII/III (2.4G2). 1x10^6 cells were stained with CD19-v421TM, CD23-APC/Cyanine7, CD5-FITC, CD39-PE/Cyanine7, CD273-APC, F4/80-PerCP/Cyanine5.5, and CD73-PE for 25 minutes on ice. Cells were washed with 2ml staining buffer, and resuspended for analysis on a LSRFortessaX20. Live cells were excluded via FSC and SSC.
Results Summary:	B1 cells and a subset of macrohpages in the peritoneum express CD73.
