Spectral cytometry revolutionized flow cytometry, enabling higher multiplexing and increasing demand for novel fluorophores and conjugates. BioLegend introduces Spark PLUS and StarBright conjugates, designed to fill spectral gaps and provide alternatives with superior performance. Here, we explore their characteristics and show improvements to an existing 39-color panel. These new conjugates can replace previously used conjugates maintaining or improving performance and resolution, enhancing data quality. Thus, our technology offers unparalleled capabilities in flow cytometry analysis.

Understanding immune cell states is paramount to the study of many of our most pressing health challenges, including cancer, infectious diseases, and aging. Profiling single cells using omics approaches has proven valuable in these contexts. While capturing single cell transcriptomic information has transformed biomedical research, here I will highlight the benefit of co-measuring protein and RNA expression together. Using our studies in different human diseases as examples, I will discuss results gained by each modality, describe our experimental workflow, and review analysis approaches.
