Optimized immunophenotyping with high dimensional spectral flow cytometry panels

To fully characterize cell function and ultimately understand pathological conditions, technologies evolve to provide more efficient ways to interrogate biological systems. One such tool that has emerged is spectral flow cytometry. With capacity to multiplex >40 parameters, the availability of so many reagents comes with challenges, making it difficult to find the best conjugates to use and increasing the complexity of the optimization process. Here we discuss BioLegend’s strategies to build complex panels, including backbone combinations that leave open channels for easy expansion. Thus, with our extensive portfolio and experience, BioLegend can help your research with either pre-optimized panels or a customized design.

Lineage-ambiguous memory T cells drive early allergic sensitization through de novo IgE generation

The pathways responsible for the emergence of IgE antibodies that drive food allergy remains poorly understood. Using a model of the early stages of food allergy, Koenig et al. found that a single gavage of allergen with cholera toxin adjuvant failed to sensitize mice, but re-exposure to the allergen alone even 1 year later caused sensitization and anaphylaxis upon allergen challenge. The first intragastric exposure induced CD44hi lineage-ambiguous CD4+ T cells but failed to activate B cells. Allergen re-exposure drove Tfh differentiation and IgE production de novo through naïve B cells dependent on IL-4 and CD40/CD40L ligation. Koenig's data implicated memory CD4+ T cells in initiating IgE production and holding IgE memory.

Streamlining single-cell isolation with bead mill dissociation

Flow cytometry, NGS, and single-cell analysis have undoubtedly advanced the field of immunology. Protocol optimization enables data reproducibility and assay consistency, critical for delicate immune cells of limiting quantities. We present method comparison work using a bead mill for sample disaggregation into single-cell suspension versus a manual, cell-strainer method in this proof-of-concept study with mouse spleen. Data is shown for cell count, cell viability, and immune cell marker populations via flow cytometry.
