Upon encountering an infection or other inflammatory stress, the hematopoietic system responds, in part, by increasing production of immune cells. Zhao et al. describe a mechanism by which hematopoietic stem and progenitor cells (HSPCs) respond to TLR stimulation and subsequent NF-κβ activation by producing diverse cytokines, surprisingly, more quickly and to a higher degree than mature immune cells. Of these cytokines, IL-6 was the most abundantly produced and this HSPC-derived IL-6 was able to stimulate myeloid cell differentiation. In addition to IL-6, neutralization of IFN-γ or GM-CSF also had an inhibitory effect on progenitor cell proliferation and survival. As HSPCs are located directly in the bone marrow stem cell niche, direct cytokine production may be advantageous to mount a rapid response.
