Recombinant human MMP1 (Cat. No. 592902; 50 ng) treated with (lane 2) and without (lane 1) active MMP3 were resolved by electrophoresis, transferred to nitrocellulose, and probed with purified monoclonal anti-MMP1 (clone M4110F03) antibody. Proteins were visualized using a goat anti-mouse-IgG secondary antibody conjugated to HRP (Cat. No. 405306) and chemiluminescence detection.

Description

Members of the matrix metalloproteinase family proteins (MMPs) are structurally related and zinc-containing enzymes that not only degrade the extracellular matrix (ECM) and connective tissue proteins under normal physiological setting but also during disease (e.g. arthritis and metastasis). The MMP family includes six different groups of enzymes: collagenases, gelatinases, stromelysins, transmembrane MMPs, matrilysins and others. MMPs are secreted as proenzymes that have to be cleaved in order to be activated. MMP1 consists of a pro-domain which is cleaved upon activation, a catalytic domain containing the zinc binding site, a short hinge region, and a carboxyl terminal (hemopexin-like repeats) domain. It is thought to play an important role in rheumatoid and osteo-arthritis pathogenesis as mediators of cartilage and bone destruction.

Substrates of MMP-1 include collagen I, II, III, VII, VIII, and X as well as casein, gelatin, alpha1 antitrypsin, myelin basic protein, L Selectin, pro-TNF, IL-1β, IGFBP3, IGFBP5, pro-MMP-2, and pro-MMP-9. In addition, chemokines are substrates for MMP-1. CCL2, CCL7, CCL8, and CCL13 are catalytically cleaved by MMP-1 to produce receptor antagonists. CXCL5 and CXCL12 are also substrates for MMP1. TIMPs inhibit MMPs in a 1:1 inhibitor to enzyme ratio through interaction of the N-terminal domain of the TIMP molecule with the active site of the MMP. MMP1 is overexpressed in invasive melanoma, colorectal, and esophageal cancers. Importantly, MMP-1 expression was shown to increase progressively with tumor stage.

Antigen Details

Structure
Monomer, belongs to the peptidase M10A family. 469 amino acids, predicted molecular weight of 54 kD.
Distribution
Extracellular. MMP-1 is expressed by fibroblasts, keratinocytes, endothelial cells, monocytes, and macrophages.
Function
MMP1 cleaves collagen of types I, II, III, VII and X. MMP-1 is thought to play an important role in the pathogenesis of rheumatoid and osteo-arthritis as mediators of cartilage and bone destruction. It is also involved in angiogenesis, tissue remodeling and cancer metastasis. MMP-1 regulates chemokine activity. MMP1 is inhibited by TIMPs, α2-macroglobulin (inhibitor of MMPs in bodily fluids). TNF-alpha; down-regulates TIMPs and enhances the expression of MMPs.
Interaction
MCP1, MCP3, MCP4, TIMP-1, MMP7, Collagen type II α1, IBP-3, Integrin α2, CD44, CD147, Brevican, Chymase 1, Aggrecan 1.
Ligand/Receptor
TIMPs.
