    Western blot of purified anti-Loricrin antibody (clone Poly19051). Lane 1: Molecular weight marker; Lane 2: 20 µg of A431 cell lysate; Lane 3: 20 µg of HepG2 cell lysate. The blot was incubated with 1 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP donkey anti-rabbit IgG antibody (Cat. No. 406401). Enhanced chemiluminescence was used as the detection system.

    Total lysates (15 µg protein) from HaCaT (human, positive control) and U87-MG (human, negative control) cells were resolved by electrophoresis (4-20% Tris-glycine gel), transferred to nitrocellulose, and probed with 1:500 (1 µg/ml) purified anti-Loricrin antibody, clone Poly19051. Proteins were visualized using chemiluminescence detection by incubation with HRP Donkey anti-Rabbit secondary antibody (Cat. No. 406401, 1:3000 dilution). Direct-Blot™ HRP anti-β-actin antibody was used as a loading control (Cat. No. 643807, 1:8000 dilution).

Description

Loricrin is a major protein component of the cornified cell envelope found in terminally differentiated epidermal cells. Mutations in this gene are associated with Vohwinkel's syndrome and progressive symmetric erythrokeratoderma, both inherited skin diseases.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemisty, a concentration range of 1.0 - 10 μg/mL is suggested. For Western blotting, a usage of 1.0 μg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
