Total cell lysates (15 µg total protein) from untransfected (-) or Cre recombinase-transfected (+) HeLa cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.1 µg/mL (1:10000 dilution) of purified anti-Cre recombinase antibody (Poly9080) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP donkey anti-rabbit IgG antibody (Cat. No. 406401) at a 1:3000 dilution. Direct-Blot™ anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:25000 dilution. Lane M: Molecular weight marker.

Description

Cre-recombinase is a 38 kD DNA recombinase derived from the P1 bacteriophage. It is highly specific for a 34 bp DNA sequence (loxP) found in P1 DNA. This polyclonal antibody was raised against a full-length Cre Recombinase recombinant protein and was subsequently purified using Protein A. It can be used to determine the success of gene manipulations in transgenic animals.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.025 - 0.5 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
