M588_Pure_CD44_Antibody_1_030719
Western blot of purified anti-CD44 antibody (clone M588). Lane 1: Molecular weight marker; Lane 2: 30 µg of human brain lysate; Lane 3: 10 µg of HeLa cell lysate; Lane 4: 30 µg of mouse Brain lysate. The blot was incubated with 5 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP-labeled goat anti-mouse IgG (Cat. No. 405306). Enhanced chemiluminescence was used as the detection system.

ICC staining of purified anti-CD44 antibody (clone M588) on HeLa cells. The cells were fixed with 4% PFA, permeabilized with a buffer containing 0.1% Triton X-100 and 0.25% BSA, and blocked with 2% normal goat serum and 0.02% BSA. The cells were then incubated with 5 µg/ml of the primary antibody for overnight at 4°C, followed by incubation with 5 µg/ml of Alexa Fluor® 647 goat anti-Rat IgG for one hour at room temperature. Nuclei were counterstained with Hoechst, and the slide was mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 40X objective. Scale bar: 50 µm

Description

CD44 is a cell-surface glycoprotein that is expressed in a large number of cell types. CD44 functions as an adhesion molecule that is important for cell migration and intercellular interactions and adhesion. CD44 transcripts undergo complex alternative splicing, which leads to a variety of functionally distinctive isoforms of this protein. Transcription of CD44 has been shows to be partially activated by Wnt/beta catenin signaling pathway, and thus is linked to tumor development.
