Good option to stain CD45.2.

Experimental Design

Application:	Flow cytometry
Cells used:	Mouse splenocytes
Brief Protocol:	Resuspend in PBS/BSA and count the cells. Load up to 5x106 cells onto a V-Bottom 96-well-plate. Spin down, resuspend in 50uL of PBS/BSA containing αFcγR (2.4G2) 30 µg/ml and incubate 15min at 4°C. Spin down and resuspend in 50 µl antibody staining solution containing anti-CD45.2 PE/Cyanine7 (1:300). Incubate 15min at 4°C in the dark. Wash twice with PBS/BSA, resuspend and transfer to FACS tubes to read. Use PI to exclude the dead cells.
Results Summary:	In C57BL/6 mice lymphocytes express the congenic marker CD45.2. Upon transfer of cells bearing CD45.1 (purified from Ly5.1+/+ mice), the use of anti-CD45.2 PE/Cyanine7 allows to identify the recipient cells CD45.2+CD45.1-.
