Bioinformatic homology analysis reveals that Oncostatin M (OSM) has a similar composition and structure as LIF, G-CSF, and IL-6; this suggests that these genes were derived by duplication of a common ancestral gene. In fact, many of its biological functions are shared with LIF. The receptor systems for OSM differ in composition between man and mouse. Rat oncostatin M receptor has 83% identity with the mouse receptor, and 58% identity with the human receptor. Rat oncostatin M has 72.1% and 51.4% identity with the mouse and human proteins respectively. Thus, it is possible the rat oncostatin M and its receptor may share more similarities with the mouse ligand –receptor system. Human OSM binds two types of OSM receptor (OSMR) complexes. The type I receptor complex is composed of gp130 (IL-6 signal transducer) and LIF receptor β (LIFRβ) subunits. The type II receptor is constituted by the gp130 receptor and the OSMR β-chain. The murine ortholog, which has high affinity binding sites, binds only to the type II OSMR/gp130 receptor complex. Human OSM efficiently activates signal transduction in murine cells; nevertheless, it utilizes only the type I LIFR/gp130 heterodimer on these cells. However, mouse OSM is unable to stimulate human cells. OSM shares the OSMRβ-chain with the IL-31 receptor complex. The OSMRβ subunit is expressed by fibroblasts, endothelial, hepatic, lung, and hematopoietic cells. Pro inflammatory properties of OSM have been reported in the skin, adipose tissue, lung, heart, and liver. OSM triggers acute phase protein synthesis on hepatocytes. Rat OSM inhibits proliferation of rat oval cells (hepatic progenitors in the adult liver, that participate in the liver regeneration) inducing differentiation to hepatocytes. Rat OSM is highly expressed in the late fetal and neonatal testis, studies in vitro suggested that rat OSM plays a role in the regulation of Leydig cell progenitor formation.Product Details

- Storage & Handling
- Unopened vial can be stored between 2°C and 8°C for one month, or at -20°C to -70°C for one year. For maximum results, quick spin vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/ml. Do not vortex. It is recommended to further dilute in a buffer (PBS, HPBS, DPBS, or EBSS) containing a carrier protein such as 0.1% BSA and stored in working aliquots at -20°C to -70°C. After reconstitution, it can be stored at -20°C to -70°C for three months. Avoid repeated freeze/thaw cycles.
- Rat Oncostatin M induces the proliferation of rat C6 glioma cells. The ED50 = 3 — 5 µg/ml.
