Whole cell extracts (15 µg protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-PDPK1 Antibody, clone W17215A, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG Antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular Weight marker.

Description

3-Phosphoinositide-Dependent Protein Kinase 1 (PDPK1) is serine/threonine kinase that plays a central role in diverse cellular pathways. It functions by phosphorylating and activating members of the AGC family of serine kinases, including Akt1, Protein Kinases A and C, and p70 ribosomal protein S6 kinase. Loss or reduction of PDPK1 in the thymus results in defective T-cell differentiation and proliferative expansion.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 1 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone is predicted to recognize rat PDPK1 due to high sequence conservation between the immunizing protein and the rat homolog.

This clone is predicted to recognize PDPK1 isoforms 1-4 due to the presence of the immunizing sequence in each isoform.

This clone failed ICC and IP testing.
