Total cell lysates (15 µg protein) from non-treated and LPS treated (1 µg/mL for 16 hours) Raw264.7 cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to nitrocellulose, and probed with 0.02 µg/mL of purified anti-Nos2 (clone W16030C) antibody and a competitor's antibody used at the manufacture's recommended concentration (upper). Proteins were visualized by chemiluminescence detection using a goat anti-rat-IgG secondary antibody conjugated to HRP for clone W16030C and a donkey anti-rabbit IgG antibody conjugated to HRP for the competitor’s antibody. Direct-Blot™ HRP anti-β-actin antibody was used as a loading control (lower). Lane M is the molecular ladder.

Non-treated (left image) and LPS treated (1 µg/mL for 16 hours, right image) Raw264.7 cells were fixed with 4% paraformaldehyde (PFA) for fifteen minutes, permeabilized with 0.5% Triton X-100 for three minutes, and blocked with 5% FBS for 60 minutes. Then the cells were intracellularly stained with 0.25 µg/mL anti-Nos2 antibody (clone W16030C) overnight at 4°C followed by Alexa Fluor® 594 (red) conjugated goat anti-rat IgG for one hour at room temperature. Nuclei were counterstained with DAPI (blue). The image was captured with a 60X objective.

Description

Nitric Oxide Synthase 2 (NOS2), also known as inducible NOS (iNOS), contains an N-terminal oxygenase domain and a C-terminal reductase domain, and functions to catalyze the formation of nitric oxide (NO) from L-arginine. NO is a reactive free radical which acts as a biologic messenger with diverse functions throughout the body, such as neurotransmission, antimicrobial, and antitumor activity. NOS2 is involved in inflammatory responses and enhances the synthesis of PGE2 and proinflammatory cytokines such as IL-6 and IL-8. The NOS2 gene is highly expressed in liver and is inducible by a combination of bacterial endotoxins and certain cytokines, including IL-1, IFNγ and TNFα. NOS2 also has nitrosylase activity and mediates cysteine S-nitrosylation of cytoplasmic target proteins such as COX2.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.002 - 0.02 µg per ml. For immunocytochemistry, a concentration range of 0.05 - 0.5 µg per ml is suggested. For flow cytometric staining, the suggested use of this reagent is ≤0.125 µg per million cells. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Mouse NOS2 has two isoforms: Isoform A is 130 kD and isoform B is 118 kD. This clone recognizes isoform B better than A in the tested cell.

Function
NOS2 is a nitric oxide synthase and produces large quantity of NO in response to the stimulation of proinflammatory cytokines. NOS2 plays an important role in host defense against pathogens and participates in anti-tumor processes.
Interaction
Forms a homodimer. Interacts with SLC9A3R1, GAPDH, S100A8, and S100A9.
