C57BL/6 mouse splenocytes were cultured for 3 days with (filled histogram) or without (dashed histogram) recombinant human TGF-β1 and recombinant mouse IL-6. Cells were removed from culture media, washed, and finally fixed and permeabilized. Cells were then intracellularly stained with purified anti-mouse AhR (clone W16012C) followed by anti-rat IgG PE.

Description

Aryl Hydrocarbon Receptor or AhR is a bHLH transcription factor that is activated by ligands such as TCDD. It is expressed in many immune cells and perform an important role in immune regulation. AhR is induced under TH17-polarizing conditions. In its inactive state, AhR presents in the cytoplasm, and upon ligand binding it translocates to the nucleus. Activation of AhR is tightly controlled by Ubiquitination, ligand metabolism, and complex disruption.

Recommended Usage

Each lot of this antibody is quality control tested by intracellular immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
