IHC staining of anti-MECP2 antibody (clone N227/21) on formalin-fixed paraffin-embedded rat brain tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., the tissue was incubated with 10 µg/ml of the primary antibody overnight at 4°C. BioLegend's Ultra Streptavidin (USA) HRP Detection Kit (Multi-Species, DAB, Cat. No. 929901) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. ICC staining of anti-MECP2 antibody (clone N227/21) on HeLa cells. The cells were fixed with methanol, permeabilized with 0.1% Triton X-100 for 20 minutes, and blocked with 2% Normal Goat Serum and 0.02% BSA for 30 minutes. The cells were then incubated with 10 µg/mL of the primary antibody overnight at 4°C, followed by incubation with 2.5 µg/mL of Alexa Fluor® 594 conjugated goat anti-mouse IgG (Cat. No. 405326) for one hour at room temperature. The cells were counterstained with DAPI. The image was captured with a 40X objective. Scale bar: 50 µm -

Methyl CpG binding protein (MECP2), also known as autism susceptibility to X-linked 2 (AUTSX3), is a nuclear protein crucial for normal neuronal function that binds to methylated DNA, forming complexes with histone deacteylases and turning off gene expression. MECP2 expression is an X-linked trait. Loss of MECP2 expression causes Rett Syndrome (also known as RTS, RTT, or RS) a neurological disorder which causes decelerated growth of hands, feet, and the head.

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.0 - 10 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
