    Total cell lysates (15 µg protein) from PC3 cells were resolved by electrophoresis (4-20% Tris-glycine gel), transferred to a nitrocellulose membrane, and probed with serial dilutions of purified anti-CD304 antibody (clone 14H4), 1:1000 (0.5 µg/mL, lane 1), 1:500 (1 µg/mL, lane 2) and 1:250 (2 µg/mL, lane 3). Proteins were visualized using chemiluminescence detection after incubation with a 1:3000 dilution of HRP conjugated goat anti-mouse-IgG secondary antibody for the anti-CD304 (upper panel) or a 1:5000 dilution of Direct-Blot™ HRP anti-β-Actin (clone 2F1-1, lower panel). Lane M: Molecular weight ladder. Non-specific bands can be detected at high antibody concentrations.

Description

The membrane isoform of CD304 is a receptor that regulates angiogenesis and organogenesis in response to growth factors of the VEGF family. CD304 forms a complex with VEGFR2/KDR, resulting in increased cell migration and chemotaxis. In the nervous system, CD304 complexed with plexin mediates Sema-3A signaling and may be involved in axon guidance.

Each lot of this antibody is quality control tested by Western blotting. Western blotting, suggested working dilution(s): Use 10 µl per 5 ml antibody dilution buffer for each mini-gel (1 µg/ml). It is recommended that the reagent be titrated for optimal performance for each application.
