Total cell lysate from HeLa cells (lane 1, 15 µg), NIH3T3 cells (lane 2, 15 µg) and mouse brain (lane 3, 15 µg) were resolved by electrophoresis (4-12% Tris-Glycine gel), transferred to nitrocellulose, and probed with purified anti-HSP60 (clone P83G8) antibody. Proteins were visualized using an HRP Goat anti-mouse IgG Antibody and chemiluminescence detection. Purified anti-β-actin antibody (poly6221) was used as a loading control.

    A549 cells were fixed with 4% paraformaldehyde (PFA) for ten minutes and then were permeabilized with 0.5% Triton X-100 for five minutes followed by blocking with 5% FBS for 30 minutes. Then the cells were intracellularly stained with 2.5 µg/mL anti-HSP60 (clone P83G8) in blocking buffer overnight at 4°C and followed by DyLight™ 594 (red) conjugated goat anti-mouse IgG for one hour at room temperature. Actin filaments were labeled with Alexa Fluor® 488 Phalloidin (green). Nuclei were counterstained with DAPI (blue). The image was captured with a 60X objective.

Description

HSP60, a mitochondrial chaperonin, is essential for the refolding, assembly, and transportation from the cytoplasm to mitochondrial matrix. It shares a high degree of homology and function similarity to bacteria GroEL. Studies also show that HSP60 may participate in some autoimmune diseases along with autosomal dominant and autosomal recessive neurodegenerative disorders.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.5 µg per ml. For immunocytochemistry, a concentration range of 1.0 - 10 μg/ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
