Whole cell extracts (15 µg protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of purified anti-eIF3F antibody (clone W19170B) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-actin antibody (Cat. No. 664804) was used as a loading control at a 1:10000 dilution (lower). Lane M: Molecular weight marker.

Description

The eukaryotic translation initiation factor 3 (eIF3) is an 800 kD oligomeric complex comprised of 13 different subunits and plays an essential role in protein translation. The eIF3 complex directly links the 43S pre-initiation complex to eIF4F-bound mRNA. The subunit eIF3F has been shown to negatively regulate both cap-dependent and IRES-dependent translation and also interacts with mTOR and S6K1, and therefore also regulates mTORC1 and mTORC2 signaling. EIF3F mRNA is downregulated in multiple types of cancer. Overexpression of eIF3F protein leads to hypertrophy by altering protein synthesis via the mTORC1 pathway. Under energy starvation conditions, eIF3F is ubiquitinated by the MAFbx/atrogen-1 complex and targeted for proteasomal degradation, leading to muscle atrophy.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.5 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone was tested for ICC using HeLa cells fixed with 4% PFA and permeabilized with either methanol or Triton X-100. Both methods resulted in poor eIF3F staining.

This clone failed to immunoprecipitate eIF3F from HeLa lysates.
