IHC staining of anti-Cytokeratin, HMW antibody (clone 34βE12) on formalin-fixed paraffin-embedded normal human skin. After antigen retrieval using Retrieve-All Antigen Unmasking System 1, the tissue was incubated with the primary antibody at 0.7 µg/ml for 60 minutes at room temperature. BioLegend's Ultra Streptavidin (USA) HRP Detection Kit was used for detection followed by hematoxylin counterstaining.

Description

This antibody is specific for high molecular weight cytokeratins 1, 5, 10 and 14. Clone 34βE12 is immunoreactive with squamous and adeno-squamous carcinomas. In normal epithelia, the antibody stains stratified epithelia, myoepithelial cells and basal cells in the prostate gland and bronchi. This antibody is also useful in differentiating benign and malignant tumors of the prostate gland.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 0.5 - 5.0 μg/ml is suggested. For Western blotting, a concentration range of 1.0 - 10 μg/ml is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

The Cytokeratin, High Molecular Weight (HMW) antibody (clone 34ßE12) reacts primarily with complex epithelium such as squamous, ductal and transitional epithelium and corresponding tumors.
