Whole cell extracts (15 µg protein) from HeLa cells transfected with non-targeting control siRNA (siCON) or siRNA targeting SHC1 (siSHC1) were resolved on a 4-12% Bis-Tris gel, transferred to PVDF and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-SHC1 Antibody, clone O91H8, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular Weight marker.

    Whole cell extracts (250 µg total protein) prepared from HeLa cells were immunoprecipitated overnight with 2.0 µg of Purified Mouse IgG1, κ Isotype Ctrl Antibody (Cat. No. 401402) or Purified anti-SHC1 Antibody, clone O91H8. The resulting IP fractions and whole cell extract input (6%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-SHC1 Antibody, clone O91H8. Description

SHC1 is a ubiquitously expressed adaptor protein involved in receptor tyrosine kinase signaling including the activation of the RAS/Raf/MAPK pathway. Three isoforms of SHC1 (p66, p52, p46) have been described, each with unique cellular functions. SHC1 has an SH2 domain and a protein tyrosine phosphate binding (PTB) domain, which recognize activated receptor tyrosine kinases. Tyrosine residues located in the CH1 domain of SHC1 are phosphorylated by tyrosine kinases and serve as docking sites for proteins such as Grb2. SHC1-bound Grb2 recruits SOS, which activates Ras, thereby promoting cell proliferation and differentiation.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 1.0 µg per ml. For immunoprecipitation, the suggested use of this reagent is 2.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.
