Chromatin Immunoprecipitations (ChIP) were performed with cross-linked chromatin samples from 4 X 106 HeLa cells with either A) 1:100 dilution of Go-ChIP-Grade™ purified anti-Histone H3 Trimethyl (Lys4) antibody, clone Poly6088, or B) equal amount of Go-ChIP-Grade™ purified rabbit polyclonal isotype control antibody, clone Poly29108 (Cat. No. 910805) by using Go-ChIP-Grade™ Protein G Enzymatic Kit (Cat. No. 699904). The enriched DNA was purified and quantified by real-time qPCR using primers targeting human GAPDH gene region and α-Satellite repeats. The amount of immunoprecipitated DNA in each sample is represented as signal relative to total amount of input chromatin.

    Specificity of Go-ChIP-Grade™ purified anti-Histone H3 Trimethyl (Lys4) antibody (clone Poly6088, Cat. No. 608803) was determined by peptide ELISA. A variety of histone modification peptides including H3K4me1, H3K4me2, H3K4me3, H3K36me1, H3K36me2 and H3K36me3 were pre-coated at 25 ng/mL and incubated with increasing concentrations of Go-ChIP-Grade™ purified anti-Histone H3 Trimethyl (Lys4) antibody. Go-ChIP-Grade™ purified anti-Histone H3 Trimethyl (Lys4) antibody specifically binds to H3K4me3 peptide in a dose-dependent manner with an ED50 of 16 ng/mL.

Description

Histone proteins are classified into core histones (H2A, H2B, H3, H4) and linker histones (H1, H5). Core histones form an octamer, which contains two H2A-H2B dimers and one H3-H4 tetramer. Core histones are predominantly globular except for the unstructured N-terminal tails. Posttranslational modifications, such as acetylation, methylation, phosphorylation, ubiquitination, SUMOylation and ADP-ribosylation occur in histone tails.

 

Histone modifications induce changes of chromatin structure and thereby affect the accessibility of transcription factors, nuclear proteins and enzymes to genomic DNA, resulting in gene activation or repression. It is known that histone modification plays critical roles in DNA repair, DNA replication, transcription regulation, alternative splicing, chromosome condensation and some diseases including autoimmune diseases and cancers.

ChIP - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by ChIP Assay. The suggested dilution for ChIP application is 1:50-1:500 by volume. It is recommended that the reagent be titrated for optimal performance before each experiment.

Application Notes

Clone Poly6088 is predicted to recognize both mouse and rat histone H3 tri-methylated at Lysine 4 due to complete sequence homology between the immunizing peptide and each murine homolog. It has displayed poor sensitivity when tested for western blot. We therefore do not recommend the clone for this application.
