ICC staining of Biotin anti-Gelsolin antibody (clone CPTC-Gelsolin-2) on HaCaT cells. The cells were fixed with 4% PFA, permeabilized with a buffer containing 0.1% Triton X-100 and 0.25% BSA, and blocked with 2% normal goat serum and 0.02% BSA. The cells were then incubated with 0.5 µg/mL primary antibody for overnight at 4°C, followed by incubation with 2.5 µg/mL Alexa Fluor® 647 Streptavidin antibody (Cat. No. 405237) for one hour at room temperature. Nuclei were counterstained with DAPI. The image was captured with a 60X objective. Scale bar: 50 µm

Description

After severing, gelsolin remains bound to the barbed end of the actin filament, preventing the reannealing of severed filaments. Gelsolin is crucial for the maintenance of actin homeostasis. Cells from gelsolin null mice exhibit a variety of defects, such as pronounced stress fibers formation, defective chemotaxis, and wound healing. Dysfunction of Gelsolin has been indicated in multiple diseases include cancer, infection, inflammation, cardiac injury, pulmonary disease, and neurodegeneration. Gelsolin is proteolytically cleaved and rendered inactive in Alzheimer’s disease (AD). Overexpression of gelsolin significantly reduces amyloid beta load in brains of AD transgenic mice.

Recommended Usage

Each lot of this antibody is quality control tested by immunocytochemistry. For immunocytochemistry, a concentration range of 0.5 - 10 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
