Experimental Design

Application:	Flow cytometry
Cells used:	Mouse splenocytes
Brief Protocol:	Mash mouse spleen through nylon mesh and use RBC lysis solution. Wash cells in FACS buffer (1x PBS+2%FBS+2mM EDTA). Resuspend cells in 100ul FACS buffer + 1ul biotin anti-mouse CD19 on ice for 10 minutes. Wash cells 2x in FACS buffer. Resuspend cells in 100ul FACS buffer + 1ul BV421-streptavidin. Wash cells 2x in FACS buffer. Fix cells overnight in 2% paraformaldehyde in PBS pH 7.4. Wash cells in FACS buffer and analyze on flow cytometer.
Results Summary:	CD19 cells are clearly labelled.
