Brief Protocol: |
Resuspend in PBS/BSA and count the cells. Load up to 5x106 cells onto a V-Bottom 96-well-plate. Spin down, resuspend in 50uL of PBS/BSA containing αFcγR (2.4G2) 30 µg/ml and incubate 15min at 4°C. Spin down and resuspend in 50 µl of antibody staining solution containing CD5BV510 (1:200). Incubate 15min at 4°C in the dark. Wash with PBS/BSA, spin down resuspend and transfer to FACS tubes to read. Use PI to exclude the dead cells. |
Results Summary: |
T cells are higher expressors of CD5, while B cells (subset of B-1a cells) that are enriched in the peritoneal cavity are intermediate expressors of CD5. |
