Anti-mouse Ki-67 APC for Flow Cytometry

Experimental Design

Application: Flow cytometry
Cells used: Mouse splenocytes
Brief Protocol: Resuspend in PBS/BSA and count the cells. Load up to 5x106 cells onto a V-Bottom 96-well-plate. Spin down, resuspend in 50uL of PBS/BSA containing αFcγR (2.4G2) 30 µg/ml and incubate 15min at 4°C. Spin down and resuspend in 50 µl antibody staining solution containing anti surface antibodies. Incubate 15min at 4°C in the dark. Wash. Fix and permeabilize the cells. Wash. Use anti-ki67 APC at (1/100) in perm/wash buffer. Incubate 30min in the dark. Wash with Perm/wash buffer. Resuspend in PBS/BSA and transfer to FACS tubes to read.
Results Summary: CD138hi cells (red) containing plasmablast and plasma cells contain higher frequency of proliferating cells (Ki67+), than B cells (blue).
