Experimental Design |
Application: | Flow cytometry |
Cells used: | Mouse splenocytes |
Brief Protocol: | Resuspend in PBS/BSA and count the cells. Load up to 5x106 cells onto a V-Bottom 96-well-plate. Spin down, resuspend in 50uL of PBS/BSA containing αFcγR (24.62) 30 µg/ml and incubate 15min at 4°C. Spin down and resuspend in 50 µl of antibody staining solution containing biotinylated antibody against the molecule to stain. Incubate 15min at 4°C in the dark. Wash with PBS/BSA, spin down and resuspend in 50 µl staining solution containing SA-Brilliant Violet 605™ (1:200 to 400). Incubate 15min at 4°C in the dark. Wash with PBS/BSA, spin down resuspend and transfer to FACS tubes to read. Use PI to exclude the dead cells. |
Results Summary: | In this case anti-mouse CD19biot was used, 50-60% of live lymphocytes should be CD19biot-SABrilliant Violet 605™+. |
Additional Notes: | As it is possible to see in the image 1/400 dilution is enough to be able to distinguish + and – populations. 1/200 dilution gives rise to a slightly brighter staining. |
