Good, reliable and not very bright.

Experimental Design

Application:	Flow cytometry
Cells used:	Mouse splenocytes
Brief Protocol:	Resuspend in PBS/BSA and count the cells. Load up to 5x106 cells onto a V-Bottom 96-well-plate. Spin down, resuspend in 50uL of PBS/BSA containing αFcγR (24.62) 30 µg/ml and incubate 15min at 4°C. Spin down and resuspend in 50 µl antibody staining solution containing anti-CD3 PerCP/Cyanine5.5 (1:100). Incubate 15min at 4°C in the dark. Wash twice with PBS/BSA, resuspend and transfer to FACS tubes to read.
Results Summary:	Approximately half of the live lymphocytes in the spleen should be CD3 PerCP/Cyanine5.5 + (T cells).
