This secondary antibody allows bright labeling of cells previously treated with biotinylated antibodies.

Experimental Design

Application:	FACS
Cells used:	in vitro derived human dendritic cells from CD14+ monocytes
Brief Protocol:	CD14+ monocytes were isolated from buffy coats and cultured for 6 days in medium, supplemented with GM-CSF and IL-4. On day 6, cells were treated with a cytokine cocktail to induce maturation of dendritic cells. On day 7, cells were stained with biotinylated anti-MHCII, followed by incubation with the BioLegend antibody Brilliant Violet 605™ streptavidin. I used 100µl of a dillution of 1µl Brilliant Violet 605™ streptavidin in 1000µl buffer to stain ~10^5 cells. Cells were analyzed using an Attune Flow Cytometer. MHCII postive cells were brightly stained.
Results Summary:	This antibody can be highly dilluted and still stain cells very brightly.
