    Stability Testing for Recombinant Mouse CD229. Recombinant mouse CD229 was aliquoted in PBS pH 7.2. One aliquot was frozen and thawed four times (4x Freeze/Thaw) and compared to the control that was kept at 4°C (Control). The samples were tested by their property to enhance the anti-CD3-induced mouse T cell proliferation in a dose-dependent manner. The ED50 for this effect is 1 – 5 µg/mL.

Description

CD229, also known as Ly-9 and SLAMF3, belongs to the SLAM family. There are two alleles in mice, Ly-9.1 and Ly-9.2. Ly-9.1 is expressed in most mice strains, while Ly-9.2 is restricted in C57BL/6 and related strains. CD229 is a type I transmembrane protein, consisting of four Ig domains in its extracellular region, a transmembrane region and a cytoplasmic region. Its N-terminal Ig domain mediates homophilic interaction and its cytoplasmic region contains consensus sequence which is responsible for binding to the SH2 domains of adaptor proteins, SAP and EAT-2. CD229 also binds to adaptor protein, Grb2 which controls CD229 internalization via AP-2-dependent clathrin-coated pits. CD229 is expressed on thymocytes, mature T and B cells. CD229 localizes at the immune synapse between T cells and B cells in an antigen-dependent manner. CD229 deficient mice show poor T cell proliferation and reduced IL-2 production upon anti-CD3 stimulation. Furthermore, those mice have mild defects in Th2 differentiation. Nevertheless, CD229 deficient mice do not present any defects in NKT cell development and macrophages function. In addition, they develop spontaneous autoimmunity and expand populations of innate memory-like CD8 T cells and invariant NKT cells in thymus. Therefore, CD229 plays a negative role in iNKT cell development. In systemic lupus erythematosus, SLAMF6 and CD229 expressions are increased in T cells, and their engagement along with antigen-mediated TCR stimulation significantly induce Th17 cell differentiation. Co-ligation of TCR and CD229 by antibodies inhibit partially the secretion of IFNγ in human peripheral blood lymphocytes.

Mouse CD229, amino acid (Lys48-Phe454) (Accession # AAH95921.1), with a C-terminal 6x His tag, was expressed in 293E cells. The carboxyl terminus contains 6His tag.
Molecular Mass
The 413 amino acid recombinant protein has a predicted molecular mass of approximately 46 kD. The DTT-reduced or non-reduced protein migrates at approximately between 62 and 80 kD by SDS-PAGE. The predicted N-terminal amino acid is Lys.

> 95%, as determined by Coomassie stained SDS-PAGE

Concentration
25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
Unopened vial can be stored between 2°C and 8°C for up to 2 weeks at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.

What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.

Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.

Does specific activity of a recombinant protein vary between lots?

Specific activity will vary for each lot and for the type of experiment that is done to validate it, but all passed lots will have activity within the established ED50 range for the product and we guarantee that our products will have lot-to-lot consistency.
