Total cell lysate from HeLa (lane 1, 15 µg), MCF-7 (lane 2, 15 µg) and Jurkat (lane 3, 15 µg) were resolved by electrophoresis (4-12 % Bis-Tris gel), transferred to nitrocellulose, and probed with purified anti-Tubulin α 8 antibody (clone P82D6D7). Proteins were visualized using a HRP goat anti-mouse-IgG secondary antibody and chemiluminescence detection. Purified anti-β-actin antibody (clone Poly6221) was used as a loading control.

    HeLa cells were fixed with 4% paraformaldehyde (PFA) for ten minutes and then were permeabilized with 0.5% Triton X-100 for five minutes followed by blocking with 5% FBS for 30 minutes. Then the cells were intracellularly stained with 2.5 µg/mL anti-Tubulin α 8 (clone P82D6D7) in blocking buffer overnight at 4°C and followed by DyLight™ 594 (red) conjugated goat anti-mouse IgG for one hour at room temperature. Nuclei were counterstained with DAPI (blue). The image was captured with a 40X objective.

Description

Tubulin, one of the major constituents of microtubules, is a dimeric protein consisting of an alpha and beta tubulin. Tubulin is a highly conserved protein that is ubiquitously expressed. Tubulin is a GTP-binding protein that can be modified by phosphorylation and acetylation. Tubulin interacts with a variety of proteins including RAC GTPase activating protein 1, ZAP70, Polo-like kinase, c-Myc, Fyn, and microtubule associated protein 1A. Tubulin α 8 belongs to the Tubulin α family which contains 8 other members such as tubulin α 1A, tubulin α 4A, tubulin α 3E. It has been shown that mutations in the tubulin α 8 gene is associated with polymicrogyria and optic nerve hypoplasia.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/ml. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Clone P82D6D7 may also recognize the other Tubulin a members since this clone was developed with partial sequence of tubulin a 8 (a.a. 294-449) that shares high percentage of homology with other members in the Tubulin a family such as tubulin a 4A, tubulin a 3C/D, and tubulin a 1A.
